Tolerance is basically how close a color has to be to the color you click to be treated as one big color (for stuff like the paint bucket and magic wand select). Eg. A tolerance of 0 selects only that exact color, while an average tolerance (about 32 [13%]) selects the exact color as well as all of the colors several shades lighter and darker.
This was made for things like photos and art that change colors ever so slightly between pixels. When spriting, it works against you so it should always be set to 0, except for extraordinary circumstances.

Just last month one of my friends showed up at my house with his deck and he's like "Ohey we haven't played Yu-Gi-Oh in forever. I suddenly need to play it." or something. So we played. He beat me twice. Then I realised "hey, this isn't my deck, this is a deck of random extra cards." so I find my real deck and proceed to curb stomp him.

You probably heard about Gliese 581d, which is 581g's neighbor.
This is relevant. A Message From Earth is basically Earth sent some messages and photos via radio waves towards Gliese 581d, so that if there is life capable of receiving radio waves, they'll learn about us. The cool thing is that Gliese 581g can catch the message as well. The Message will get there in 2029.
