I know it's possible, but I can't figure how to make a kill all enemy-triggered goal/door/pipe. Can anyone tell me how?

Is it possible to make Yoshi's House invisible until activated by an event?O.O
If I understand what you want correctly, you may want to tinker around with the No sprite on screen = Goal sprite.

Well, that's not exactly what I had in mind. What I had in mind was that the level automatically finishes after removing all sprites, or a door/pipe appears. I don't really have enough hacking knowledge to "tinker" with this No Sprite On Screen = Goal sprite :O

And the key/keyhole thing wouldn't work with a door/pipe anyway.

Did you actually try inserting this?
It was just a modified version of mikeyk's, still having comments on it from the original.
It does this. Alternatively you can use the level ender sprite included with spritetool.

You have to overwrite one of the "revealed" level tile graphics with the yoshi's house one.
To spawn a pipe when all sprites on screen are killed, you might want to edit the custom sprite called "door generator" It explains how to change what map16 block in spawns, so you could just change the spawning of the top part of a door to the left part of a pipe, then the bottom part to another section of pipe, then make another copy of the sprite for the right sides (Unless you can figure out how to do it all in one.)
Two more sprites for horizontal pipes.

If I remember correctly, there's just a sprite that spawns a pipe once all sprites are dead, isn't there?
Thanks guys. I've figured this out now. I'll just leave this thread up for the Invisible Yoshi's House. :D
