Is it possible to create custom bosses without using an already existing sprite and adding custom animation?
Of course as a custom boss is still a custom sprite. There is a tutorial about custom sprites (albeit not finished as it's missing the custom interaction and the boss part).

I know programming with Java, how easy would a transition of Java skills to ASM be?

It would be as if you never knew Java at all. Really.

I mean, having some coding experience might help in giving you a basic understanding in the way that programming languages function in general, but ASM is a much lower level of code than Java or C or anything like that. It's a pretty different story.
Not to mention that you should start with easier stuff at first anyway.
